Glutathione: The Master Antioxidant

Glutathione (gamma-L-glutamyl-L-cysteinyl-glycine; GSH) is a tripeptide present in virtually every mammalian cell, serving as the body’s primary intracellular antioxidant defense. IV vs. Oral Bioavailability: The Case for Intravenous Administration

The bioavailability argument for IV glutathione is compelling and well-documented. Witschi et al. (1992) conducted a landmark pharmacokinetic study demonstrating that oral glutathione supplementation at doses up to 3,000mg produced no statistically significant increase in circulating glutathione levels, plasma cysteine, or glutathione in erythrocytes. This is because intact glutathione is rapidly hydrolyzed by gamma-glutamyltranspeptidase (GGT) in the intestinal brush border and hepatic first-pass metabolism further reduces bioavailability to an estimated 3-5%. Intravenous administration bypasses both barriers entirely, delivering 100% of the administered dose to systemic circulation. Allen and Bradley (2011) confirmed that IV glutathione produces immediate and dose-dependent increases in plasma GSH levels, with peak concentrations achieved within minutes of infusion completion and elevated levels sustained for 2-4 hours post-infusion.

Melanogenesis Modulation: How Glutathione Brightens Skin

Glutathione’s skin-brightening mechanism operates through several complementary pathways in melanocyte biology. First, glutathione directly inhibits tyrosinase — the rate-limiting enzyme in melanin synthesis — by chelating copper ions at the enzyme’s active site (Villarama and Maibach, 2005). Second, glutathione shifts melanogenesis from eumelanin (brown/black pigment) toward pheomelanin (yellow/red pigment) by conjugating with L-DOPA, diverting the biosynthetic pathway away from the eumelanin branch. Third, glutathione’s antioxidant activity reduces oxidative stress in melanocytes, which is a known trigger for melanogenesis upregulation. Watanabe et al.
